Capivasertib is a targeted cancer medicine you take by mouth. It is used with fulvestrant to treat certain breast cancers. Your care team will confirm if this drug fits your cancer type and explain how it will be given as part of your treatment plan.

Warning(s)

  • High blood sugar can happen and may be severe. Watch for confusion, unusual thirst or hunger, peeing more often, fast breathing, or breath that smells fruity. If you have diabetes, you may need closer blood sugar checks. Tell your care team about any changes.
  • Serious skin reactions can occur. Get help right away for a new or worsening rash; red, peeling, or very dry skin; blisters on the lips, skin, eyes, or mouth; or fever, chills, sore throat, shortness of breath, swollen glands, or severe tiredness.
  • Kidney problems and dehydration can happen, especially if you have severe diarrhea. Call your care team if you cannot pass urine, have a big change in urine amount, blood in urine, or sudden weight gain.
  • Infections, including urinary tract infections (UTIs), can occur. Call your care team for fever, burning or pain with urination, pelvic or lower belly pain, or blood in urine.
  • Electrolyte changes may cause muscle cramps, weakness, shaking, loss of balance, seizures, loss of appetite, severe nausea/vomiting, or an abnormal heartbeat. Get medical help if these happen.
  • Chest pain or pressure needs urgent medical evaluation.
  • Allergic reactions can be serious. Seek help for rash; hives; itching; wheezing; chest or throat tightness; trouble breathing or swallowing; or swelling of the face, lips, tongue, or throat.
  • Drug interactions are common. Some HIV medicines, antifungals, certain antibiotics, seizure medicines, and herbal products like St. John’s wort may interact. Do not start, stop, or change any medicine or supplement unless your care team says it is safe.
  • Avoid grapefruit and grapefruit juice. They can change how the drug is processed in your body.
  • Monitoring is important. You may need regular blood tests to check blood sugar, electrolytes, kidney function, and triglycerides. Follow your care team’s schedule.
  • Pregnancy and birth control: This drug may harm an unborn baby. You will have a pregnancy test before starting. If you can become pregnant, use effective birth control while taking this drug and for 1 month after the last dose. If your sex partner can become pregnant, use birth control during treatment and for 4 months after the last dose. If a pregnancy occurs, call your care team right away.
  • Do not breastfeed while taking this drug.
  • If you are 65 or older, you may have a higher chance of side effects. Report new or worsening symptoms promptly.
  • Tell all of your health care providers that you take capivasertib, including doctors, nurses, pharmacists, and dentists.

Kind of Cancers It Is Used For (not a complete list)

  • Hormone receptor (HR)-positive, HER2-negative breast cancer that is locally advanced or metastatic.
  • It is used in people whose tumors have certain gene changes in the AKT pathway (such as PIK3CA, AKT1, or PTEN). Your tumor may be tested to see if this drug is likely to help.
  • Capivasertib is taken together with fulvestrant.

Not all breast cancers are the same. Your oncologist will use your cancer’s biomarkers and prior treatments to decide if capivasertib is appropriate for you.

Mechanism of Action

Capivasertib is a targeted therapy called an AKT inhibitor. AKT is a protein that helps control cell growth and survival in the PI3K/AKT pathway. Many cancers use this pathway to grow and resist treatment.

  • Capivasertib blocks AKT (AKT1/AKT2/AKT3), turning down signals that tell cancer cells to grow and divide.
  • By slowing this pathway, capivasertib can slow tumor growth and may help your hormone therapy (fulvestrant) work better.
  • Because AKT also affects how your body handles sugar, blocking AKT can raise blood sugar levels.

Common Side Effects

  • Diarrhea (common). Severe diarrhea can lead to dehydration. Drink fluids as advised. Tell your care team if diarrhea is severe, keeps coming back, or you feel lightheaded or weak.
  • Upset stomach, nausea, or vomiting.
  • Decreased appetite.
  • Change in taste.
  • Feeling tired or weak.
  • Mouth irritation or mouth sores.
  • Skin changes, including rash or redness/irritation on the palms of the hands or soles of the feet.
  • High blood sugar.
  • High triglycerides on blood tests.
